This review article is dedicated to ether-lipids that are forming a class of anti-cancer agents and that can interact with plasma membrane leading to trigger apoptosis or to reduce cancer cell migration. In this article a first part is dedicated to the pro apoptotic Edelfosine and to its mechanism of action that starts from interactions with the plasma membrane to trigger apoptosis. The second part is focused on Ohmline (a more recently discovered molecule - 2011) which is an efficient and selective modulator of SK3 ion channel. This action, can inhibit the formation of bone metastasis in a murine model of metastatic breast cancer. In this article, a structure-activity relationship is presented for both compounds (Edelfosine and Ohmline) and in terms of mechanism of action, a reorganization of lipid raft appeared as an essential step.
